Evaluation of autoantibody profiles in a cohort of new-onset pediatric systemic lupus erythematosus patients using autoantigen microarray technology

چکیده
Purpose Systemic lupus erythematosus (SLE) is characterized by the production of autoantibodies directed against many highly-conserved nuclear antigens. 15% of SLE patients develop disease in childhood or adolescence, and pediatric (pSLE) patients often have more severe disease onset and organ system involvement. Antigen microarray technology allows the comprehensive analysis of autoantibodies directed against hundreds of antigens with minimal amounts of sera. The purpose of this study was to characterize autoantibody profiles in a prospective cohort of new-onset pSLE patients at onset and within 6-9 months of disease onset and to determine if autoantibody profiles differentiate patients with distinct clinical manifestations such as nephritis.

Autoantigen microarrays reveal autoantibodies associated with proliferative nephritis and active disease in pediatric systemic lupus erythematosus
INTRODUCTION Pediatric systemic lupus erythematosus (pSLE) patients often initially present with more active and severe disease than adults, including a higher frequency of lupus nephritis. Specific autoantibodies, including anti-C1q, anti-DNA and anti-alpha-actinin, have been associated with kidney involvement in SLE, and DNA antibodies are capable of initiating early-stage lupus nephritis in ...

High-throughput methods for measuring autoantibodies in systemic lupus erythematosus and other autoimmune diseases.
Numerous groups have now validated high-throughput approaches to autoantibody profiling in a variety of systems. Recently, we have used autoantigen microarray technology to identify distinct autoantibody profiles in H-2 congenic MRL/lpr mice (Sekine et al., manuscript in preparation), and we are expanding this platform to study human and mouse models of IDDM and RA. We are also developing prote...
متن کاملAutoantigen Microarray for High-throughput Autoantibody Profiling in Systemic Lupus Erythematosus
Systemic lupus erythematosus (SLE) is a complex autoimmune disease characterized by the production of autoantibodies to a broad range of self-antigens. Profiling the autoantibody repertoire using array-based technology has emerged as a powerful tool for the identification of biomarkers in SLE and other autoimmune diseases. Proteomic microarray has the capacity to hold large number of self-antig...
متن کاملMeasurement of autoantibodies in pediatric- and adolescent-onset systemic lupus erythematosus and their significant relationship with disease-associated manifestations
Purpose Pediatric-onset systemic lupus erythematosus (SLE) patients are prone to develop major organ involvement and a more severe disease course than adult-onset SLE. Early management of the disease is necessary to prevent further complications, which can partly be accomplished by monitoring autoantibody levels and understanding their significance in the disease process. We evaluated an autoan...
